摘  要:将鼠抗Cd^2+-IEDTA单克隆抗体和鼠抗Pb^2+-IEDTA单克隆抗体分别与包载CdSe/CdS量子点聚苯乙烯微球偶联,建立一种可快速检测铅镉的量子点荧光定量串联卡及免疫层析竞争检测方法。串联卡的最佳反应时间为5 min,可检测的Pb^2+和Cd^2+质量浓度范围分别为1~500 ng/mL和1~400 ng/mL,检测变异系数小于10%;检测特异性良好,与其他金属离子无明显交叉反应;利用该方法与电感耦合等离子体-质谱(inductively coupled plasma-massspectrometry,ICP-MS)法进行肉类食品检测对比,检测结果与ICP-MS相关性良好。该检测方法为同时定量检测铅镉提供快速简便的有效途径,适用于监测肉中Pb2+和Cd2+含量是否超标。The objective of this study was to establish a quantum dot-based fluorescence immunochromatographic assay for the rapid simultaneous detection of lead and cadmium.Mouse anti-Cd^2+-IEDTA and anti-Pb^2+-IEDTA monoclonal antibodies were separately coupled with CdSe/CdS core-shell quantum dots embedded in polystyrene microspheres.The optimum reaction time of the dual test strip was 5 min.Pb^2+concentrations of 1–500 ng/mL,and Cd^2+concentrations of 1–400 ng/mL were detectable using the proposed method with coefficients of variation of less than 10%.Moreover,there was no crossreactivity with other metal ions.The results of this assay were correlated well with those of inductively coupled plasma-mass spectrometry(ICP-MS)for commercial meat samples.This dual fluorescence immunochromatographic assay provided an applicable method for the rapid and simultaneous quantitative detection of lead and cadmium in meat.
