摘  要:建立了高效液相色谱法测定多类食品中胭脂虫红的含量。试样用盐酸于水浴酸化后,用甲醇溶液提取,考察了不同用量的溶剂对提取结果的影响,选择用2mol/L盐酸溶液10.0mL和甲醇15.0mL作为提取液。提取液经聚酰胺固相萃取小柱净化后,用高效液相色谱(带二极管阵列检测器)进行检测,外标法定量。样品检测采用TC-C18(4.6mm×150mm,5μm)色谱柱,流动相为0.05%甲酸水溶液(A)和乙腈(B),流速为1.0mL/min,梯度洗脱,检测波长为276nm。结果表明,在2.5~100mg/L范围内线性关系良好,相关系数r=0.99998,方法的检出限为1.25mg/kg。不同浓度的加标水平下,测定平均回收率在96%~105%之间,相对标准偏差(n=6)为0.1%~0.9%。该方法操作简便、灵敏度高、检测范围广,适用于各类食品中胭脂虫红含量的检测。A method for determination of carmine cochineal in foods by high performance liquid chromatography (HPLC) was developed in this paper.The sample was extracted with hydrochloric acid-methanol and then purified by SPE column,finally detected by high performance liquid chromatography( HPLC) with diode array detector( DAD), the quantitative analysis was performed by external standard method.Carminic acid was conducted on a C18 column (4.6 mm × 150 mm,5 μm) with a mobile phase consisted of 0.05% formic acid and acetonitrile under a gradient elution condition at a flow rate of 1.0 mL/min.This method used for determing carminic acid at 276 nm.There was a good linear relationship in the range of 2.5~100 mg/L,with linear correlation coefficient r=0.99998.The detection limit was 1.25 mg/kg.The recovery of standard addition was between 96%~105%.The relative standard deviation(n=6) was 0.1%~0.9%.This method was proved to be fast、sensitive、reliable and good linearity with low relative standard deviation,which can be suitable for the determination of carmine cochineal in various kinds of food.
