摘  要:从碱蓬根际土壤中分离筛选获得一株能产吲哚乙酸(IAA)的促生菌株BG-5,对该菌株进行了16S rDNA序列分析。以IAA的产量作为评价指标,利用单因素试验及响应面分析法对发酵条件及培养基成分进行优化。结果表明,BG-5菌株被鉴定为短小芽孢杆菌(Bacillus pumilus),最佳培养条件为装液量30 mL/250 mL、初始pH值为8、培养温度37℃;优化后的培养基成分为酵母提取物7.3 g/L,胰蛋白胨8.8 g/L,七水硫酸镁7.2 g/L;在此最优条件下,BG-5菌株的吲哚乙酸产量为87.86μg/mL,约为优化前的2倍。A growth-promoting and indoleacetic acid(IAA)-producing strain BG-5 was isolated and screened from the rhizosphere soil of Suaeda salsa,and analyzed by 16S rDNA sequences.Using the yield of IAA as evaluation index,the fermentation conditions and medium composition were optimized by single factor tests and response surface methodology.The results showed that the strain BG-5 was identified as Bacillus pumilus.The optimal culture conditions were loading volume 30 ml/250 ml,initial pH 8 and culture temperature 37℃.The optimal medium composition was yeast extract 7.3 g/L,tryptone 8.8 g/L,and magnesium sulfate heptahydrate 7.2 g/L.Under the optimal conditions,the IAA yield of the strain BG-5 was 87.86μg/ml,which was about twice that of before optimization.
