摘  要:利用碱性蛋白酶酶解松仁清蛋白制备高抗氧化活性肽,通过超滤、SephadexG-25、SephadexG-15及反相高效液相色谱对抗氧化肽进行分离纯化,并采用电喷雾串联质谱进行结构鉴定。经筛选获得高抗氧化活性肽苯丙氨酸-苯丙氨酸-脯氨酸-酪氨酸(FFPY)和酪氨酸-亮氨酸-脯氨酸-苯丙氨酸(YLPF),分子质量分别为572.67 Da和538.65 Da。对FFPY和YLPF进行固相合成,纯度达到99.16%和99.91%,氧自由基吸收能力分别为7 329.63 μmol/g和2 835.47 μmol/g。Two peptides with strong antioxidant activity were seprate and purified from the alkaline protease of pine nut kernel albumin hydrolysate by by ultrafiltration, column chromatographies on Sephadex G-25, and Sephadex G-15 and reversed-phase high-performance liquid chromatography, and their structure was identified by electrospray ionization tandem mass spectrometry (ESI-MS/MS). The peptides were found to be Phe-Phe-Pro-Tyr (FFPY) and Tyr-Leu-Pro-Phe (YLPF) with molecular masses were 572.67 and 538.65 Da, respectively. Solid phase synthesis of FFPY and YLPF was performed with a purity of 99.16% and 99.91%. It was proved that the oxygen radical absorbance capacity (ORAC) values of FFPY and YLPF were 7 329.63 and 2 835.47 μmol/g, respectively.
