摘　　要：Bafilomycin A1是大环内酯类抗生素,具有广泛的抗癌活性。通过HPLC及HPLC-MS分析Streptomyces bacillaris ATCC 15855^T的发酵产物,结果显示S.bacillaris ATCC 15855 T发酵能够产生bafilomycin A1。S.bacillaris ATCC 15855^T分离自中亚土壤。通过全基因组测序,获得了S.bacillaris ATCC 15855^T的完整基因组序列。该基因组由6957417 bp的线性染色体组成,GC含量为72.29%。该染色体预测了7284个开放阅读框架(ORF),18个rRNA操纵子,64个tRNA基因。通过antiSMASH v 4.0.3分析,共预测了39个生物合成基因簇。其中,具有抑制液泡型H(+)ATP酶的bafilomycin A1基因簇与已报道的基因簇相似度为100%。在bafilomycin A1合成基因簇中有19个ORFs。Bafilomycin A1 is a kind of macrolide antibiotic with extensive antitumor activity.Analyzing the fermentation product of Streptomyces bacillaris ATCC 15855^T by HPLC,the result showed that S.bacillaris ATCC 15855^T is a producer of bafilomycin A1.S.bacillaris ATCC 15855^T was isolated from soil sample derived from Central Asia,and the complete genome sequence of S.bacillaris ATCC 15855^T was obtained,which consisted of a linear chromosome of 6957417 bp with a GC content of 72.29%.The chromosome contained 7284 predicted Open Reading Frames(ORFs),18 rRNA operons and 64 tRNA genes.S.bacillaris ATCC 15855^T was a relatively novel Streptomyces species proved by phylogenetic analysis.Thirty-nine biosynthetic gene clusters were predicted by the analysis of antiSMASH v 4.0.3.Among them,bafilomycin A1,a kind of antibiotic with inhibiting vacuolar-type H(+)-ATPase,showed similarity of 100%to reported gene clusters.There were 19 ORFs in bafilomycin A1 biosythentic gene cluster.
