摘  要:建立一种快速准确的柱前衍生-超高效液相色谱法,同时测定黄酒中17种游离氨基酸含量。黄酒样以6-氨基喹啉-N-羟基琥珀酰亚胺基氨基甲酸酯(AQC)衍生化处理,以流速为0.7 mL/min经Waters AccQ·TagTM Ultra C18(1.7μm,2.1×100 mm)的色谱柱梯度洗脱,利用二极管阵列检测器,在260 nm波长处检测。结果显示,17种游离氨基酸浓度在25~400μmol/L范围内,峰面积与浓度之间具有良好的线性关系(R2≥0.9993),回收率在94.21%~104.82%。该方法可在11 min内将17种游离氨基酸完全分离,具有准确、快速、重复性好的特点,可用于黄酒中游离氨基酸含量的测定。A rapid method of UPLC coupled with pre-column derivatization has been developed for simultaneous determination of 17 kinds of free amino acids in Huangjiu.Wine samples were derived with 6-aminoquinolyl-N-hydroxysuccinimidylcarbamate(AQC)as derivatization agent,then separated on a Waters AccQ·TagTM Ultra C18 column(1.7μm,2.1×100 mm)at a flow rate of 0.7 mL/min,and finally detected by using PDA detector at the wavelength of 260 nm(UV).The results showed that,the concentration of the 17 amino acids had good correlation(R2≥0.9993)with peak areas in the range of 25μmol/L to 400μmol/L,and the spike recoveries were between 94.21%to 104.82%.All the 17 free amino acids could be separated completely within 11 min by use of this method.The method was featured by accurate and rapid determination with good repeatability.
