摘 要:为了实现高效生物合成鸢尾酮,开展从陈化鸢尾根状茎中分离霉菌并进行生物合成条件的优化研究。首先从鸢尾根状茎中分离微生物,并结合HPLC分析,分离到一株产鸢尾酮的菌株。随后对发酵产物进行分离纯化,并经过液质和核磁氢谱结构验证,确证发酵产物中含有cis-α-鸢尾酮和cis-γ-鸢尾酮。然后通过形态及系统进化树分析,确定分离得到的菌株为烟曲霉。最后对发酵温度、初始pH值、氮源和反应时间进行优化,获得了比较适合的生物合成条件(30℃、初始pH值6.5和10 g/L尿素)。在这一条件下,经12 d的发酵培养,鸢尾酮产量达到7.25 mg/L,相当于从1 kg干鸢尾根状茎中获得了145 mg鸢尾酮。这一工作对微生物发酵法规模化生产鸢尾酮具有一定的参考意义。In order to produce irones efficiently,the culture condition of one new strain isolated from aged iris rhizome was optimized.One new strain ZD-J1 was isolated and irones could be produced by analyzing the fermentation broth with HPLC.The structure of irones were further identified by using HPLC/MS and 1 H NMR which contained cis-α-irone and cis-γ-irone.Then,the strain ZD-J1 was confirmed to be aspergillus fumigates according to morphological characterizations and phylogenetic treeanalysis.Further optimization of fermentation conditions were carried out,and the suitable conditions were determined(30℃,initial pH 6.5 and 10 g/L urea).Under these conditions,the mass concentration of total irones was improved to 7.25 mg/L after 12 d fermentation,which corresponding to up to 145 mg/kg of irones from raw dried rhizomes.This work will be helpful to the large-scale production of irones by microbial transformation.
