摘 要:以国内外10个不同品种紫甘薯为研究对象,测定其干物率和花色苷含量,评价不同品种花色苷的体外抗氧化能力.结果表明,不同紫甘薯花色苷粗提液体外抗氧化能力存在差异,其中徐紫薯8号总抗氧化能力最强,为(90.87±2.84)U/mL.进一步采用阳离子交换树脂纯化徐紫薯8号花色苷粗提液,并测定其花色苷主要成分含量.结果显示,经纯化后的提取液中花色苷质量分数达到21.95%.建立高脂诱导肥胖小鼠模型,灌胃纯化后的徐紫薯8号花色苷提取液(800 mg/(kg·d))进行干预,9周后检测其对小鼠肝脏中丙二醛(MDA)、超氧化物歧化酶(SOD)、过氧化氢酶(CAT)和谷胱甘肽过氧化物酶(GSH-Px)含量的影响.结果表明,花色苷干预的高脂组小鼠MDA含量显著低于高脂模型组,而SOD,CAT和GSH-Px含量均显著高于高脂模型组,说明紫甘薯花色苷对高脂饮食小鼠具有较强的抗氧化能力.The contents of dry matter and anthocyanin in 10 purple sweetpotato(PSP)cultivars were tested,and different varieties of PSP anthocyanin(PSPA)rough extract in vitro antioxidant capacities were determined aiming to find out the best purple sweetpotato cultivar.The contents of anthocyanin and the antioxidant activities from different varieties were different,and Xuzi 8 exhibited the highest antioxidant capacity.Xuzi 8 anthocyanin rough extract was purified by cation exchange resin,and the main components in refined anthocyanin were determined.The anthocyanin concentration of refined extract was 21.95%.Eight-week-old male ICR strain mice were fed on either a normal diet,or a high fat diet(HFD)and intragastrically administered refined anthocyanin 800 mg/kg once a day.The content of malonaldehyde(MDA),superoxide dismutase(SOD),catalase(CAT)and glutathione peroxidase(GSH-Px)in mice were detected after 9 weeks.The results showed that the MDA content in liver of HFD+PSPA group mice was lower than that of HFD group mice,and the content of SOD,CAT and GSH-Px in liver of the HFD/PSPA group mice were significantly higher than that of HFD group mice.Studies indicate that the refined anthocyanin have strong antioxidation effect on high-fat diet mice.
