摘  要:建立一种缓解视疲劳片中全反式叶黄素的含量测定方法。样品经水超声破膜后用0.1%二丁基羟基甲苯(BHT)乙醇溶液超声提取,高效液相色谱法测定,采用YMC Carotenoid C30 (4.6 mm×250 mm, 5μm)为色谱柱;以甲醇/水(88︰12, V/V)为流动相A相,甲基叔丁基醚为B相,两相均含0.1%BHT,梯度洗脱,流速0.8 mL·min^-1,检测波长445 nm,柱温30℃,检出限9.1×10^-6 mg·kg^-1。结果表明,全反式叶黄素在0.94~9.44μg·mL^-1浓度范围内线性关系良好,相关系数r=0.999 9;加标浓度0.272 7 mg·mL^-1,平均回收率100.48%, RSD=1.59%(N=9)。该方法专属性、精密度、准确性、重复性好,操作简便,计算简单,适用于缓解视疲劳类产品中全反式叶黄素的含量测定,为有效控制该类产品的质量提供依据。Aim to establish a method for the content determination of trans-lutein for relieving visual fatigue. The samples were ultraphoniced after splitted micro-capsules, and extracted by 0.1% butylated hydroxytoluene(BHT) ethanol and analyzed by HPLC. The samples were separated on YMC carotenoid C30(4.6 mm×250 mm, 5 μm) column, gradient eluted with A-phase mixture of methanol and water(88︰12, V/V), B-phase methyl tertiary butyl ether, both phases containing 0.1% BHT;The flow rate was 0.8 mL·min^-1, the detection wavelength was 445 nm, and oven temperature was 30 ℃;Detection limit was 9.1×10^-6 mg·kg^-1. The results showed that the trans-lutein had a good linear within the range of 0.94-9.44 μg·mL^-1, correlation coefficient of 0.999 9, the adding standard concentration of 0.272 7 mg·mL^-1, the average recovery of 100.48%, and the RSD of 1.59%(N=9). The method was specific, accurate, repeatable, efficient and easy to operate and calculate, and could be used for quantitative analysis of trans-lutein for relieving visual fatigue. The basis for controlling the quality of the kind of products effectively was provided.
