摘 要:提取葛根多酚氧化酶(Ployphenol Oxidase,PPO)的粗酶,研究不同浓度的水杨酸、苹果酸、酒石酸、抗坏血酸和柠檬酸处理和联合两个不同有机酸处理对葛根PPO相对活性及其护色效应的影响。结果表明,5种有机酸对葛根PPO都有抑制效果,且随有机酸浓度增高抑制效果增强。在有机酸浓度达到60 mmol·L^-1时,酒石酸的抑制效果最好,其相对酶活仅为9.67%,抗坏血酸和水杨酸的抑制效果不显著,其相对酶活为80.33%和78.67%。联合两种有机酸处理样品出现不同类型的效应,其中苹果酸与水杨酸联合处理表现出最强的加和效应;而柠檬酸、酒石酸分别与水杨酸共同处理时,加和效应、次加和效应、协同效应均有体现。抗坏血酸对葛根泥的护色效果显著,而对抑制酶活有限,同一浓度下水杨酸则对葛根泥无护色效果。The crude enzyme of polyphenol oxidase(PPO)from kudzu root was extracted.The effect of five organic acids(alicylic acid,malic acid,tartaric acid,ascorbic acid and citric acid)and combined with two different organic acids on the polyphenol oxidase activity and color protection of kudzu root was analyzed.The results showed that the inhibitory effects of five organic acids on kudzu root PPO increased with the increase of organic acid concentration.The inhibition effect of tartaric acid was the best with a concentration of 60 mmol·L^-1,and its relative enzyme activity was 9.67%,and the inhibition effect of ascorbic acid and salicylic acid was not significant,their relative enzyme activity was 80.33%and 78.67%.The combination treatment of two organic acids showed different inhibition types to kudzu root PPO.The salicylic acid with malic acid showed the strongest additive effect,while both the salicylic acid with citric acid and tartaric acid showed additive effect,subadditive effect and synergistic effect.Ascorbic acid had a best color protection effect on kudzu root pulp,while it showed a limited inhibition to kudzu root PPO.And salicylic acid had no color protection effect to kudzu root pulp at the same concentration.
