摘　　要：为得到植物乳杆菌的最优发酵培养基,研究了不同碳源、氮源对植物乳杆菌活菌数的影响,应用Box-Behnken实验设计和响应面法对葡萄糖、酵母浸粉、牛肉膏的质量浓度进行了优化,利用1 L发酵罐对发酵条件进行研究,最终得到植物乳杆菌BC00171摇瓶的最优培养基配方为葡萄糖20 g/L、酵母浸粉30 g/L、硫酸镁0.3 g/L、醋酸钠7 g/L、柠檬酸三铵2 g/L、磷酸氢二钾2 g/L、硫酸锰0.3 g/L、吐温801 mL/L,在此培养基配方下发酵20 h后活菌数达到3.9×10^9 CFU/mL。发酵罐最优培养基配方及发酵工艺优化结果是:葡萄糖40 g/L、酵母浸粉30 g/L、硫酸镁0.3 g/L、醋酸钠7 g/L、柠檬酸三铵2 g/L、磷酸氢二钾2 g/L、硫酸锰0.3 g/L、吐温801 mL/L,按照接种量3%,pH调控至6.8,培养12 h活菌数即可达到3×10^9 CFU/mL。In order to obtain the optimal fermentation medium of Lactobacillus plantarum,the experiment studied the effects of different carbon and nitrogen sources on the viable number of Lactobacillus plantarum bacteria.Box-behnken experimental design and response surface method were used to optimize the concentration of glucose,yeast extract powder and beef paste.Finally,the optimal medium formula of Lactobacillus plantarum BC00171 in shake flask was obtained(glucose 20 g/L,yeast extract powder 30 g/L,magnesium sulfate 0.3 g/L,sodium acetate 7 g/L,triammonium citrate 2 g/L,dipotassium phosphate 2 g/L,manganese sulfate 0.3 g/L,and tween 801 mL/L).After fermentation of 20 h in this medium,the viable number of bacteria reached 3.9×10^9 CFU/mL.The optimal culture medium formula in fermentor process is as follow:glucose 40 g/L,yeast extract powder 30 g/L,magnesium sulfate 0.3 g/L,sodium acetate 7 g/L,triammonium citrate 2 g/L,dipotassium phosphate 2 g/L,manganese sulfate 0.3 g/L,and tween-801 mL/L.According to the inoculum 3%,pH 6.8,culture 12 h,the living number of bacterium could reach 3×10^9 CFU/mL.
