摘  要:以自主分离的冠突散囊菌菌(Eurotium cristatum)SL-1为研究对象,对该菌株的固态发酵条件及培养基配方进行了研究。结果表明,以夏秋茶叶作为唯一氮源及发酵基质,SL-1菌株产孢的最佳培养基配方为1%(m/V)葡萄糖+1%(m/V)蔗糖,并添加由KH2PO4+K2HPO4、ZnCl2、MgSO4、CaCl2、MnSO4、FeCl3组成,终浓度为0.5%的无机盐。最适发酵条件如下:最适温度为25℃,最适初始pH为5.0,最适初始含水量为75%,最适接种量为105 CFU/g,最适发酵时间为7 d。在最佳培养基和培养条件下,获得SL-1菌株的最高产孢量(8.25×10^9 CFU/g),可为后续发酵工艺研究提供数据支撑。Using E.cristatum SL-1 self-isolated as research object,the fermentation conditions and the formula of culture medium were optimized.The results showed that the best medium by using tea-leaf as the only nitrogen source and matrix of summer-autumn tea were as follows:1%glucose(m/V)+1%sucrose(m/V),and adding inorganic salt(composed with KH2PO4+K 2HPO4,ZnCl2,MgSO4,CaCl2,MnSO4,FeCl3)with the final concentration of 0.5%.The optimal fermentation conditions were as follows:culture temperature 25℃,initial pH 5.0,initial water content 75%,inoculation amount of 105 CFU/mL and fermentation for 7 days.Under the optimal medium and culture conditions,the number of viable bacteria reached 8.25×10^9 CFU/g,which could meet the needs of the subsequent fermentation process.
