摘  要:目的:探究不同存储条件对太行菊多糖的抗氧化性及稳定性的影响。方法:采取体外检测法对比太行菊多糖对DPPH自由基的清除能力,分析不同储存条件对其抗氧化性及稳定性的影响。结果:太行菊多糖清除DPPH自由基的IC50为0.54 μg/mL,小于阳性对照组Vc的IC50;相对于碱性条件,酸性条件能更好地保持太行菊多糖清除DPPH自由基的能力;随温度的升高,太行菊多糖对DPPH清除率和抗氧化活性逐步降低;随光照时间的增加,太行菊多糖对DPPH自由基清除率小幅度降低,耐光性较好;柠檬酸和苯甲酸钠对太行菊多糖的抗氧化活性和稳定性影响较大;金属离子会影响太行菊多糖的稳定性,Al^3+、Cu^2+、Fe^3+离子溶液中太行菊多糖清除DPPH自由基的能力下降幅度较大,抗氧化活性下降趋势明显。结论:太行菊多糖的还原力较强;在低温、光照时间短、钠离子条件下能正常储存,酸性条件能增加太行菊对DPPH自由基清除率;含有Al^3+、Cu^2+、Fe^3+金属离子、柠檬酸、苯甲酸钠溶液和高温会影响多糖的稳定性,对DPPH自由基清除率和抗氧化活性都在降低,所以储存或运输太行菊多糖尽量避免和含有金属离子、柠檬酸和苯甲酸钠类物质直接接触。Objective:To investigate the effect of different storage conditions on antioxidant activity and stability of polysaccharides from Opisthopappus taihangensis.Methods:The in vitro detection method was used to analyze the DPPH scavenging ability of polysaccharides from Opisthopappus taihangensis,and the effect of different storage conditions on its antioxidant capacity and stability was analyzed.Results:The IC50 of DPPH free radical scavenging by polysaccharide was 0.54 μg/mL,which was less than the IC50 of Vc in the positive control group.Compared with alkaline conditions,acidic conditions can better maintain the ability of polysaccharides to scavenge DPPH free radicals.With the increase of temperature,the clearance of DPPH and antioxidant activity of polysaccharides from Opisthopappus taihangensis gradually decreased.With the increase of the light time,the DPPH clearance of for polysaccharides from Opisthopappus taihangensis decreased slightly,and the light resistance was better.Citric acid and sodium benzoate had a significant effect on the antioxidant activity and stability of polysaccharides from Opisthopappus taihangensis.Most of the metal ions affected the stability of polysaccharides from Opisthopappus taihangensis,and the ability of polysaccharides to remove DPPH free radicals in Al^3+,Cu^2+,and Fe^3+ metal ion solutions decreased greatly,and the antioxidant activity declined significantly.Conclusion:The reducing power of polysaccharide from Opisthopappus taihangensis was very strong.Under the condition of low temperature,short light time and sodium ion,it can be stored normally and increase DPPH clearance rate under strong acidic conditions.Containing AI^3+,Cu^2+,Fe^3+ metal ions,citric acid,sodium benzoate solution and high temperature affected the stability of polysaccharides,DPPH clearance rate and antioxidant activity were reduced,polysaccharides from Opisthopappus taihangensis should be stored or transported to avoid direct contact with metal ions,citric acid and sodium benzoate.
