摘  要:采用聚乙二醇(PEG)-硫酸铵双水相体系(ATPS)将香菇柄水提液中的多糖与蛋白质进行分离萃取研究,得香菇柄蛋白富集于上相,糖类富集于下相。综合考察PEG 4000、(NH4)2SO4、Na Cl浓度及p H对萃取结果的影响,通过正交试验进一步优化试验条件,得出最佳萃取参数。结果表明,香菇柄水提液5.00 mL, PEG 4000浓度18%,硫酸铵浓度17%, NaCl浓度1%, pH 6.5时,蛋白质萃取率达81.57%。Separation and extraction of polysaccharides and proteins from the water extract of Lentinus edodes stalk by polyethylene glycol(PEG)-ammonium sulfate aqueous two-phase system(ATPS). Lentinus edodes stem protein was enriched in the upper phase and sugar was enriched in the lower phase was studied. The effects of PEG 4000,(NH4)2SO4, NaCl concentration and pH on the extraction results were comprehensively investigated. The orthogonal experiment was used to optimize the experimental condition, and the optimum extraction parameters were obtained. The results showed that when the concentration of PEG 4000 of 18%, the concentration of ammonium sulfate of 17%, the NaCl concentration of 1%, and the pH of 6.5, the extraction rate of protein reached 81.57%.
