Optimization of Extraction Process of Proanthocyanidins from ′Cabernet Sauvignon′ Residue of Ningxia by Response Surface Methodology

摘  要:以乙醇作为提取剂,通过添加纤维素酶和果胶酶提取宁夏‘赤霞珠’葡萄皮渣中原花青素,并在单因素的试验基础上采用响应面法研究料液比、复合酶添加量、提取温度和提取时间对原花青素得率的影响,然后建立二次多项式回归模型,分析和预测原花青素的得率。试验所得最佳工艺条件为:乙醇浓度60%,复合酶添加量0.4%,料液比1∶20,pH值4.5,提取温度60℃,提取时间80 min,此条件下宁夏‘赤霞珠’葡萄皮渣原花青素的含量为22.914 mg/g。Ethanol was used to extract the proanthocyanidins from Ningxia ’ Cabernet Sauvignon’ grape skin residue by adding cellulase and pectinase. Based on the single-factor experiment, the response surface methodology was used to investigate the effects of material-liqiud ratio, the amount of complex enzyme, extraction temperature, and extraction time on the yield of proanthocyanidins. Then,the quadratic polynomial regression model was established to analyze and predict the yield of proanthocyanidins. The best process conditions for proanthocyanidins are ethanol concentration 60%,compound enzyme addition0. 4%,material-liqiud ratio 1∶20,pH value 4. 5,extraction temperature 60 ℃,extraction time 80 min. Results show that the yield of proanthocyanidins from Ningxia ’ Cabernet Sauvignon’ grape skin residue is 22. 914 mg/g.
