摘  要:为制备具有抗痛风活性的金枪鱼蛋白肽,本研究以金枪鱼为研究对象,以蛋白回收率及XOD抑制率为指标,通过单因素实验选择中性蛋白酶和胰蛋白酶为复合酶,经响应面优化得到复合酶最佳酶解条件为:复合酶解温度45.5℃,加酶量935 U·g^-1,复合酶解时间6.5 h;对于蛋白回收率,三因素影响顺序为:加酶量>复合酶解温度>复合酶解时间;对于XOD抑制率,三因素影响顺序为:复合酶解温度>复合酶解时间>加酶量。此条件下进行酶解,测得蛋白回收率为83.46%,与预测值偏差1.2%,比单一蛋白酶解提高36.1%;XOD抑制率为43.51%,与预测值偏差0.8%,比单一蛋白酶解提高54.4%,表明复合酶解后效果提高显著。此时酶解液主要含小分子肽,(1~3)KDa肽段含量为42.57%,<1 KDa肽段含量为35.98%,大分子肽含量相比于未酶解时降低了90.7%。研究结果表明该复合酶法制取的金枪鱼抗痛风活性肽具有高蛋白回收率及良好的XOD抑制率,该技术对金枪鱼抗痛风活性肽的进一步深入研究和应用具有一定的理论指导意义。To prepare tuna protein peptides with anti-gout activity,this study took tuna as the research object,using protein recovery and XOD inhibition rate as indicators,through single factor experiment,neutral protease and trypsin were selected as complex enzymes,and the optimal enzymatic hydrolysis conditions of complex enzymes after response surface optimization were:Compound enzymolysis temperature was 45.5℃,enzyme amount was 935 U·g^-1,compound enzymolysis time is 6.5 h;for the protein recovery rate,the sequence of the three factors was as follows:the amount of enzyme added>composite digestion temperature>composite digestion time;for the XOD inhibition rate,the sequence of the three factors was as follows:complex enzymolysis temperature>complex enzymolysis time>enzyme addition amount.Under the conditions of enzymatic hydrolysis,the measured protein recovery rate was 83.46%,a deviation of 1.2%from the predicted value,which was 36.1%higher than that of single proteolysis;the XOD inhibition rate was 43.51%,a deviation of 0.8%from the predicted value,and an increase of 54.4%compared with single proteolysis,the results showed that the effect of complex enzymolysis was significantly improved.At this time,the enzymolysis solution mainly contained small molecule peptides,the content of(1-3)KDa peptides was 42.57%,and the content of<1 KDa peptides was 35.98%,and the content of macromolecular peptides was reduced by 90.7%compared with that without enzyme digestion.The research results showed that the tuna anti-gout active peptide prepared by the composite enzymatic method had high protein recovery and good XOD inhibition rate.This technology had certain theoretical guiding significance for the further in-depth study and application of the tuna anti-gout active peptide.
