摘  要:建立高效液相色谱法测定多类食品中胭脂虫红的含量。试样用盐酸于水浴中酸化后,用甲醇溶液提取,根据不同类型的产品,提取液经聚酰胺固相萃取小柱净化或不净化后,用高效液相色谱—带二极管阵列检测器进行检测,外标法定量。研究在276nm和494nm波长下的检测结果,两者均在2.5~100mg/L范围内呈良好的线性关系,相对标准偏差分别为0.49%和0.76%,加标回收率分别为97%~102%和96%~102%。对8种类型的样品进行对比分析,使用净化法,两种波长可用于各类食品检测;直接提取法,在276 nm下,所有样品均无法定量,在494nm下,含干扰物质较少的样品可定量分析。A method for determination of carminic cochineal in foods by high performance liquid chromatography(HPLC)was developed in this paper.The sample was acidified with hydrochloric acid in water bath,extracted with methanol and then purified and non-purified by SPE column according to the type of samples,finally detected by HPLC with diode array detector(DAD),The quantitative analysis was performed by external standard method.With wavelength 276nm and 494nm,both determination results had good linear relationship in the range of 2.5~100mg/L.The relative standard deviations were 0.49%and 0.76%.The recoveries of standard addition were 97%~102%and 96%~102%.We also made quantitative analysis of 8 kinds of samples,both wavelengths can be used for quantitative analysis of Carmine cochineal in various kinds of food by purification method.In direct extraction method,with wavelength 276 nm,all samples cannot be quantified,with wavelength 494 nm,samples with less interfering substances can be quantitatively analyzed.
