摘 要:捕光复合物蛋白(LHCP)在植物光合作用过程中发挥重要的作用。基于水稻基因组信息,以亚洲栽培稻籼稻品种黄华占、长雄蕊野生稻及其杂种F1为试验材料,克隆了水稻Lhcb2基因,并对其基因内含子信息、蛋白序列特征、进化树、蛋白结构、基因表达进行分析。结果表明:水稻Lhcb2基因全长880 bp,含有1个长98 bp的内含子。开放阅读框长度为792 bp,编码263个氨基酸;水稻LHCB2与13个其他植物的LHCB氨基酸序列一致性为68.78%。进化树分析显示,水稻LHCB2蛋白与同属禾本科的毛竹和麻竹亲缘关系最近,与低等植物团藻亲缘关系最远。水稻LHCB2蛋白属于亲水性蛋白,其理论分子量为28 495.40,理论等电点为5.62,具有2个无序化区域,无序化比例为30.04%;水稻LHCB2蛋白三级结构有2个β折叠和3个α螺旋组成。Lhcb2基因荧光定量分析结果显示,长雄蕊野生稻和杂交F1代植株叶片中表达量分别为黄华占的1.22和1.96倍。长雄蕊野生稻中的叶绿素a和b、色素、类胡萝卜素等均高于黄华占及其杂交F1代。长雄蕊野生稻叶绿素a的含量分别为另2份水稻材料的1.13和1.75倍,叶绿素b含量为1.10和1.60倍,色素含量则为1.13和1.72倍。Light-harvesting complex protein(LHCP) plays an important role in plant photosynthesis. In this study, the Lhcb2 gene was cloned from of Oryza sativa(Huanghuazhan) and O.longistaminata based on the genome database. Its gene sequences, intro sequences, protein characteristics, phylogenetic tree, protein structure, gene expression patterns were analyzed. Sequence analysis indicated that the length of Lhcb2 gene of rice was 880 bp with a 98 bp intron, encoding 263 amino acids. Multiple alignments of LHCB with related 13 plant species indicated that the identity of them was 68.78 %. Phylogenetic tree analysis showed that the LHCB2 of rice was closely related to Dendrocalamus latiflorus and Phyllostachys edulis, but far from Volvox carteri. Protein analysis suggested that the LHCB2 of rice is a hydrophilic protein with the theoretical relative molecular weight 28495.40 and the theoretical isoelectric point 5.62. LHCB2 protein had 2 disordered regions with disorder percent 30.04%. The three-dimensional structures prediction showed that LHCB2 contains 3α-helix and 2 extended strands.The results of quantitative real time PCR showed that the expression levels of Lhcb2 gene in leaves of in‘Huanghuazhan’and F1 were 1.22 and 1.96 times than that of O.longistaminata,respectively.The contents of chlorophyll a,chlorophyll b,pigment and carotenoid in O.longistaminata were higher than that in‘Huanghuazhan’and F1.The contents of chlorophyll a,chlorophyll b and pigment of O.longistaminata were 1.13 and1.75,1.10 and 1.60,1.13 and 1.72 times than that in in‘Huanghuazhan’and F1,respectively.
