摘 要:试验旨在探究添加富硒酵母对黄曲霉毒素(aatoxin B1,AFB1)中毒雏鸡的脾脏中IL-2、IFN-γ、IL-6、TNF-α水平变化,从而探究富硒酵母对AFB1中毒雏鸡免疫功能的影响。试验选择60只1日雏鸡,随机分为3组。对照组基础日粮硒含量为0.4 mg/kg,AFB1组为基础日粮添加0.3 mg/kg AFB1,富硒酵母组添加量为0.5 mg/kg。用原子荧光吸收法检测脾脏中硒含量。用ELISA和qRT-PCR两种试验方法检测各组脾脏组织中IL-2、IFN-γ、IL-6、TNF-α的含量。结果显示:饲粮中添加AFB1降低脾脏中硒含量(P<0.05),添加富硒酵母可逆转AFB1造成的硒含量降低。qRT-PCR结果显示,AFB1组脾脏中IL-2 mRNA、IFN-γmRNA、IL-6 mRNA极显著降低(P<0.01),TNF-αmRNA的含量显著低(P<0.05);富硒酵母组脾脏中IL-2 mRNA、IL-6 mRNA显著降低(P<0.05)。ELISA结果显示,AFB1组IL-2、IFN-γ、TNF-α含量显著降低(P<0.05);富硒酵母组IL-6显著降低(P<0.05)。富硒酵母组TNF-α极显著降低(P<0.01),AFB1可导致雏鸡脾脏中IL-2、IL-6、TNF-α和IFN-γ的含量和mRNA转录水平不同程度降低,含AFB1日粮中添加富硒酵母后,可通过升高脾脏中IL-2、IFN-γ、IL-6、TNF-α含量减轻AFB1导致的免疫抑制。The aim of the study was to investigate the changes of IL-2,IFN-γ,IL-6,TNF-αlevels in the spleen of chickens poisoned with Aflatoxin B1(AFB1)by Se-enriched yeast,and to explore the effect of Se-enriched yeast on the immune function of chickens poisoned with AFB1.Sixty one-day-old chicks were randomly divided into three groups.The content of selenium in basal diet was 0.4 mg/kg in control group,0.3 mg/kg AFB1 in AFB1 group and 0.5 mg/kg in Seenriched yeast group.Determination of selenium in spleen by atomic fluorescence spectrometry.The contents of IL-2,IFN-γ,IL-6 and TNF-αwere detected by ELISA and qRT-PCR.The results showed that the content of selenium in spleen decreased with the addition of AFB1 in diet(P<0.05),and the decrease of selenium caused by AFB1 could be reversed with the addition of Se-enriched yeast.The results of qRT-PCR showed that IL-2 mRNA,IFN-γmRNA,IL-6 mRNA in spleen of AFB1 group were significantly decreased(P<0.01),TNF-γmRNA was significantly decreased(P<0.05),and IL-2 mRNA and IL-6 mRNA in spleen of Se-enriched yeast group were significantly decreased(P<0.05).The levels of IL-2,IFN-γ,TNF-αin AFB1 group were significantly decreased(P<0.05),IL-6 in Se-enriched yeast group were significantly decreased(P<0.05),and TNF-αin Se-enriched yeast group were extremely decreased(P<0.01).AFB1 can decrease the concentration of IL-2,IL-6,TNF-αand IFN-γin the spleen of chicks and mRNA transcription level,the immunosuppressant induced by AFB1 could be alleviated by increasing the contents of IL-2,IFN-γ,IL-6 and TNF-αin the spleen.
