摘 要:以乙醇为溶剂提取滁菊多酚,选择乙醇体积分数、料液比、提取时间和提取温度进行单因素试验,通过响应面试验得到滁菊多酚提取的最佳工艺条件为:乙醇体积分数64%、液料比22 mL/g、提取时间90 min、提取温度70 ℃,滁菊多酚实际提取率为32.13 mg/g。通过HPLC-QQQ/MS对滁菊粗提液多酚进行检测,得到13个响应较好的色谱峰,对其中的5个峰进行了鉴定,分别为绿原酸、异绿原酸A、木犀草苷以及2种二咖啡酰奎宁酸,其中异绿原酸A是滁菊含量最为丰富的多酚化合物。因此可以大量制备滁菊多酚并为滁菊多酚的分离纯化及生物活性研究奠定基础。Polyphenols were extracted by ethanol solvent from Chrysanthemum morifolium cv.Chuju.Four factors including ethanol volume fraction,solvent/material ratio,extraction time and extraction temperature were selected for the single-factor experiment.The best extraction conditions for extracting polyphenols in Chrysanthemum were confirmed by response surface analysis as follow:ethanol volume fraction was 64%,solvent/material ratio was 22 mL/g,extraction te was 90 min,extraction temperature was 70 ℃.Under these conditions,the actual extraction rate of polyphenols was 32.13 mg/g.HPLC-QQQ/MS analysis showed that the polyphenols in Chrysanthemum could be well analyzed.Thirteen peaks were found and five (Chlorogenic acid,isochlorogenic acid A,luteolin and two kinds of dicaffeoylquinic acids) of them were identified.Iso-chlorogenic acid A was the most abundant polyphenol in Chrysanthemum.So it was possible to prepare a large quantity of polyphenols in Chrysanthemum and to lay a foundation for the separation and purification of polyphenols in Chrysanthemum and the study of their biological activities.
