摘 要:建立了气相色谱-串联质谱技术(GC-MS/MS)同时测定山药酒中4种植物甾醇及胆固醇含量的方法,并以1,1-二苯基-2-三硝基苯肼(DPPH)自由基清除率为指标评价了山药酒体外抗氧化能力。样品经皂化、提取、浓缩后用正已烷定容,衍生化后上机分析,内标法定量。经方法学验证,各甾醇类化合物定量限为0.78~48.29μg/L,标准曲线线性关系良好(R2≥0.991),加标回收率为98.74%~101.3%,相对标准偏差(RSD)小于1.2%。采用该方法对三类市售山药酒进行分析,结果表明其甾醇类化合物组成相近,但含量存在显著差异,其中β-谷甾醇在各山药酒中含量均较高,其次是菜油甾醇,对其进行抗氧化评价,山药蒸馏酒和发酵酒的抗氧化能力强于山药露酒,为山药酒企业保健指标的测定及保健功效提供数据支撑。A method for simultaneous determination of 4 phytosterols and cholesterol in yam alcoholic drinks was established by GC-MS/MS.At the same time,the DPPH free radical scavenging rate was determined to evaluate the in vitro antioxidant ability of yam alcoholic drinks.After saponification,extraction and concentration,the sample was quantified with n-hexane,after derivatization,the sample was analyzed and quantified by internal standard method.By methodology validation,the quantization limits of each sample were between 0.78μg/L and 48.29μg/L,while calibration curves showed good linearity(R2≥0.991).The average recoveries ranged from 98.74%to 101.3%with the relative standard deviations(RSD)less than 1.2%.The three kinds of yam wine were analyzed by the method.Results showed the composition of sterols was similar,but the content was different.The content ofβ-sitosterol was high in all yam alcoholic drinks,followed by campesterol.The antioxidant activity of distilled yam alcoholic drinks and fermented yam wine were higher than blended yam wine,which provided data support for the determination of health indicators of yam alcoholic drinks enterprises and health care effects.
