摘  要:为寻求苯酚-硫酸法进行佛手瓜水溶性多糖含量测定的最优条件,探讨了试剂添加顺序、试剂用量、显色测定条件等因素对吸光度值的影响,优化因素并进行精密度、稳定性、回收率的研究.结果表明:试剂最佳添加顺序为多糖溶液,苯酚,浓硫酸;试剂最佳用量为浓硫酸5.5 mL,苯酚0.9 mL;显色测定最佳条件为温度40℃,时间5 min;最大吸收波长485 nm.优化后的苯酚-硫酸法是测定佛手瓜中水溶性多糖含量的一种高稳定性和高回收率的方法,对将来利用其中活性物质进行佛手瓜精深加工具有参考价值.In order to find the optimal conditions for the determination of water-soluble polysaccharide content in the chayote by phenol-sulfuric acid method,for the first time,this paper discussed the effect of various factors including reagent adding sequence,reagent dosage,and coloring conditions on the determination results and optimized these factors.The precision,stability and recovery of the phenol-sulfuric acid method was also studied.The results showed that the best adding sequence of the reagent was polysaccharide solution-phenolconcentrated sulfuric acid;the best reagent dosage was concentrated sulfuric acid 5.5 mL,phenol 0.9 mL;the best color development conditions were temperature 40℃,time 5 min;the max absorbance wavelength was 485 nm.Phenol-sulfuric acid method was a high stability and high recovery method for the determination of water-soluble polysaccharide content in the chayote and laid a foundation for the future deep processing of chayote using active substances.
