摘 要:目的采用正交试验法优化紫山药中花青素的提取工艺。方法采用加热提取法,以花青素含量为评价指标,采用单因素试验及L9(34)正交试验考察固液比、提取温度、提取时间、盐酸浓度因素对花青素含量的影响。结果影响提取工艺的因素依次为:料液比>温度>盐酸浓度>提取时间。紫山药中花青素的最佳提取工艺为:在料液比1∶10条件下,于2% HCl-40%乙醇溶剂中60 ℃提取40 min。结论优化的提取工艺简单、合理、可行,可用于紫山药中花青素的提取。Objective To optimize the extraction technic of anthocyanins from Purple Dioscorea alata L.by orthogonal test method.Methods To use heating extraction method and take anthocyanin content as the evaluation index,to investigate the effects of solid-liquid ratio,extraction temperature,extraction time and hydrochloric acid concentration on anthocyanin content with single factor test and L9(34)orthogonal test.Results The factors affecting the extraction process were as follows:solid liquid ratio>temperature>hydrochloric acid concentration>extraction time.The optimum extraction parameters of anthocyanins in purple Dioscorea alata L.were as follows:60℃,2%HCl-40%ethanol,extract for 40 min,the solid-liquid ratio is 1:10.Conclusion The optimized extraction process is simple,reasonable and feasible,which can be used for the extraction of anthocyanins from purple Dioscorea alata L..
