摘 要:从上海浦东新区新场镇某农场种植田筛选出1株高产α-环糊精葡萄糖基转移酶的菌株,初步研究其酶学性质,以获得有良好热稳定性和pH稳定性的酶。通过形态学特征、生理生化试验鉴定和16S rRNA序列比对分析,确定该菌株为浸麻类芽孢杆菌(Paenibacillus macerans),命名为P.macerans TLLY7。酶学性质测定结果表明,该酶的酶活性为1.35 U/mL,其最适反应温度为50℃、最适反应pH值为6.0,并且在40~45℃、pH 6.0~9.0稳定性良好。Ca^(2+)、Mn^(2+)和Zn^(2+)对酶表现出促进作用,Fe^(2+)、Cu^(2+)和Co^(2+)则对该酶表现出抑制作用。其中Ca^(2+)对酶的促进作用最强,酶活性促进率为25%,Cu^(2+)对酶的抑制作用最强,酶活性抑制率为33%。K^(+)、Mg^(2+)对酶活性的影响较小。菌株TLLY7所产α-环糊精葡萄糖基转移酶的酶学性质初步研究表明其具有深入研究的价值和应用的潜力。A high-yieldα-cyclodextrin glycosyltransferase strain was screened from planting fields on a farm in Xinchang town,Pudong New Area,Shanghai,and its enzymatic properties were studied to improve the thermal stability and pH stability of this enzyme.According to the morphological characteristics,physiological and biochemical identification and 16S rRNA sequence alignment,the strain was identified as Paenibacillus macerans,and was named P.macerans TLLY7.Enzymatic properties indicated that 50℃and pH 6.0 were the most suitable reaction conditions.It showed good stability under the conditions of 40-45℃and pH 6.0-9.0.Ca^(2+),Mn^(2+)and Zn^(2+)had positive effects on the enzyme,while Fe^(2+),Cu^(2+)and Co^(2+)had negative effects.Ca^(2+)had the strongest positive effect on enzyme activity with an increase of 25%,Cu^(2+)had the strongest negative effect with an inhibition rate of 33%.The effect of K^(+)and Mg^(2+)on the enzyme activity was not obvious.Preliminary studies on the enzymatic properties ofα-cyclodextrin glucosyltransferase produced by strain TLLY7 indicate that it has the value of further research and potential application.
