摘  要:建立快速、准确测定蛋糕、面包及饼干中甜蜜素的双波长瑞利光散射方法。在pH 9.27的碱性Tris-盐酸介质中,甜蜜素与灿烂绿以静电作用相互反应生成离子缔合物,使瑞利光散射显著增强,并产生具有2个强特征散射峰的新瑞利散射光谱,最大瑞利散射峰位于341 nm,次大瑞利散射峰位于470 nm,在这2个波长处,甜蜜素质量浓度在0.004~0.50 mg/L范围内与体系的瑞利光散射增强强度(ΔI_(RLS))呈良好的线性关系,检出限分别为0.0036 mg/L(341 nm)和0.0040 mg/L(470 nm)。当用双波长(341 nm+470 nm)瑞利光散射法测定时,检出限为0.0019 mg/L,定量限分别为1.05 mg/100 g(蛋糕)、0.974 mg/100 g(饼干)和0.994 mg/100 g(面包),样品的加标回收率为97.3%~103%,相对标准偏差(n=5)为1.1%~2.6%。该法简便、快速、灵敏,用于实际样品分析,结果满意。A novel dual-wavelength Rayleigh light scattering(DWO-RLS)method for fast and accurate determination of the food additive cyclamate in cake,bread and biscuit was established.In the alkaline Tris-hydrochloric acid medium at pH 9.27,cyclamate reacted with brilliant green via electrostatic interaction to form an ionic association complex,leading to a distinctly enhanced RLS of the system,and producing a new RLS spectrum with two strong characteristic scattering peaks.The strong peak was located at 341 nm and the weak peak at 470 nm.There was a good linear relationship between the magnitude of enhanced RLS intensity(ΔI_(RLS))and cyclamate concentration in the range of 0.004 to 0.50 mg/L.The limits of detection(LOD)were 0.0036 and 0.0040 mg/L at 341 and 470 nm,respectively.The limits of quantitation(LOQ)of the DWO-RLS method were 1.05,0.974 and 0.994 mg/100 g for cake,biscuit and bread,respectively.The recoveries of spiked samples were in the range from 97.3%to 103%,and the relative standard deviations(RSD,n=5)ranged from 1.1%to 2.6%.The method was simple,rapid and highly sensitive,and it was applied to actual samples analysis with satisfactory results.
