摘 要:目的探索中国传统中药细柱五加叶水提取物降血糖和抗氧化作用。方法采用MTT法测定细柱五加叶水提取物细胞毒活性;运用体外酶学试验,以阿卡波糖和熊果酸作为阳性对照,测定其α-葡萄糖苷酶、α-淀粉酶、基因重组人蛋白酪氨酸磷酸酶PTP1b的抑制活性评估其降血糖活性;以抗坏血酸为阳性对照,运用1,1-二苯基-2-三硝基苯肼(DPPH)自由基,2,2-联氮-二(3-乙基-苯并噻唑-6-磺酸)二铵盐(ABTS)自由基清除法来研究其体外抗氧化活性。结果细柱五加叶水提取物无细胞毒性,其体外酶抑制活性α-葡萄糖苷酶[IC_(50)(138.50±1.21)μg·mL^(-1)],α-淀粉酶[IC_(50)(914.90±0.35)μg·mL^(-1)]和PTP1B[IC_(50)(106.60±0.33)μg·mL^(-1)]。DPPH和ABTS自由基清除活性的IC_(50)值分别为(72.96±0.72)μg·mL^(-1)和(74.86±0.33)μg·mL^(-1)。结论细柱五加叶水提物具有降糖和抗氧化作用。Objective To explore the water extract of the leaves of the Chinese Pharmacopoeia records of traditional Chinese medicine Acanthopanax gracilistylus possess excellent hypoglycemic properties and antioxidant effects. Methods Using in vitro enzymology experiments, using acarbose and ursolic acid as positive controls, hypoglycemic activities were assessed based on the inhibitory activities with α-glucosidase, α-amylase, and PTP1 B;Ascorbic acid was used as a positive control, using 1,1-diphenyl-2-trinitrophenylhydrazine(DPPH) free radical, 2,2-diazo-bis(3-the free radical scavenging method of ethyl-benzothiazole-6-sulfonic acid) diammonium salt(ABTS) was used to study the in vitro antioxidant activity of the water extract of the leaves of Acanthopanax gracilistylus. Results In vitro enzyme inhibitory activity of α-glucosidase(IC_(50) 138.50 ± 1.21 μmol·L-1), α-amylase(IC_(50) 914.90 ± 0.35 μmol·L-1) and PTP1 B(IC_(50) 106.60 ± 0.33 μmol·L-1) in water extracts from the leaves of Acanthopanax gracilistylus. The IC_(50) values of DPPH and ABTS free radical scavenging activities were 72.96 ± 0.72 μg·mL^(-1) and 74.86 ± 0.33 μg·mL^(-1), respectively. Conclusion It is reported for the first time here that the water extract of the leaves of Acanthopanax gracilistylus has hypoglycemic and antioxidant effects.
