摘  要:建立固相萃取-高效液相色谱法检测高油脂植物源性食品中纽甜含量的方法。样品中纽甜经甲醇水溶液超声提取后,采用PRiME MCX固相萃取柱净化处理,经Atlantis T3色谱柱分离,以乙腈-0.1%磷酸溶液为流动相,进行梯度洗脱,采用二极管阵列检测器进行检测,外标法定量。结果表明,纽甜标准溶液在0.5~100μg/mL质量浓度范围内线性良好,线性相关系数为0.999 9,检出限为0.06 mg/kg,定量限为0.2 mg/kg。在不同种类样品中开展0.2, 2和10mg/kg这3个水平的加标回收试验,试验方法的平均回收率在90.0%~96.0%,相对标准偏差(n=6)在0.9%~1.9%,该方法前处理操作简便,具有较高的灵敏度和准确性,适用于高油脂植物源性食品中纽甜的定量分析。A method for the determination of neotame in oil-rich plant-derived food by solid phase extraction combined with high performance liquid chromatography was established. The neotame in samples was extracted by ultrasound with methanol aqueous solution, and then purified by PRiME MCX SPE column. A gradient elution was carried out on Atlantis T3 column with acetonitrile and 0.1% phosphoric acid aqueous solution as mobile phases. The target compound was detected by a diode array detector, and the quantitative analysis was done by external standard method. The results showed that the linearity of neotame was satisfied in the concentration range of 0.5-100 μg/mL with the good correlation coefficient(R2=0.999 9). The limit of detection was 0.06 mg/kg, and the limit of quantitative was 0.2 mg/kg. The average recovery rates at three spiked levels of 0.2, 2 and 10 mg/kg ranged from 90.0% to 96.0%, with relative standard deviations(n=6) of 0.9%-1.9%. The method was proved to be efficient, sensitive and accurate, and it was suitable for quantitative analysis of neotame in oil-rich plant-derived food.
