摘  要:通过在平板和固体培养2种培养基质中添加外源硒并研究其对云耳(Auricularia auricula-judae)菌株百云6号菌丝生长的影响。试验结果表明,菌株百云6号菌丝体具有很强的耐硒能力,于2种培养基中表现出来的硒敏感度及忍耐力差别较大。硒浓度≤35.44 mg·L^(-1)(平板法)和硒浓度≤66.44 mg·kg^(-1)(固体法)对菌丝生长均有促进作用;硒浓度≥44.29 mg·L^(-1)(平板法)和硒浓度≥88.58 mg·kg^(-1)(固体法)对菌丝生长有抑制作用。硒浓度达到442.90 mg·L^(-1)(平板法)或442.90 mg·kg^(-1)(固体法)时,平板培养法的菌丝生长已受到严重抑制,抑制率达到69.06%;而固体培养法的抑制率只有9.52%,但菌丝体的生理变化却非常明显,主要表现为不能扭结形成耳芽,即不能由营养生长转化为生殖生长,此外条状菌丝形态明显且数量较多。The test by adding exogenous sel enium to either plating culture medium or solid culture medium showed that the mycelium of Auricularia auricula-judae Baiyun No.6 strain had strong selenium tolerance,and the selenium sensitivity and tolerance in the two culture media were quite different.The selenium concentration≤35.44 mg·L^(-1)(plate method)and 66.44 mg·kg^(-1)(solid method)both had a positive effect on mycelial growth,the selenium concentration≥442.90 mg·L^(-1)(plate method)and 88.58 mg·kg^(-1)(solid method)had an inhibitory effect on mycelial growth,and the selenium concentration≥442.90 mg·L^(-1)(plate method)and 88.58 mg·kg^(-1)(solid method)had an inhibitory effect on mycelial growth.With the selenium concentration of 442.90 mg·L^(-1)(plate method)or 442.90 mg·kg^(-1)(solid method),plate culture method of mycelial growth has been severely inhibited,and the inhibition rate was 69.06%,while the solid culture method of inhibition rate was only 9.52%,but the mycelium physiological changes were very obvious,mainly the mycelium can’t be twisted to form ear buds,and mycelium was no longer able to convert from nutritional growth to reproductive growth.In addition,the streaks of mycelium were very distinct and numerous.
