摘  要:利用常压室温等离子体(ARTP)诱变仪对产高温淀粉酶重组毕赤酵母进行诱变处理,并通过台盼蓝平板显色法初筛出产酶优势菌株,再通过摇瓶复筛选育出一株高温淀粉酶高产菌株A28,其酶活力达到1165 U/mL,较初始菌株酶活力提高37%。并对该菌株的发酵周期、接种量、诱导温度及甲醇添加量进行优化,得到最佳发酵时长为7 d、接种量为8%、诱导温度为26 ℃、甲醇添加量为1%/d,在该条件下,发酵酶活达到1835 U/mL。在连续10代的传代培养中,其酶活力保持在1830 U/mL左右,具有重要的工业应用价值。The high-temperature amylase-producing recombinant Pichia pastoris was mutagenized with an atmospheric pressure room temperature plasma (ARTP) mutagenizer,and the dominant enzyme-producing strains were initially screened by the Trypan blue plate color method,and then re-screened by shaking flasks.A high-temperature amylase high-yielding strain A28 was bred,and its enzyme activity reached 1165 U/mL,which was 37% higher than the initial strain.And fermentation period of the strain,induced temperature,inoculation quantity,and the amount of methanol added were optimized.The best fermentation time was 7 days,the inoculum was 8%,the induction temperature was 26 ℃,and the methanol addition was 1%/d.Under these conditions,the fermentation enzyme activity reached 1835 U/mL.In the subculture for 10 consecutive generations,its enzyme activity remained at about 1830 U/mL,It has important industrial application value.
