摘 要:建立了一种高效液相色谱法(HPLC)定量测定胞苷、鸟苷、胞嘧啶、尿嘧啶、次黄嘌呤、腺苷、尿苷、胸苷、腺嘌呤、虫草素的方法。在蛹虫草子实体人工栽培过程中,以不添加植物激素培养的蛹虫草为对照组,将向培养基中按照6、12、24 mg/kg的比例分别添加萘乙酸和6-苄氨基嘌呤(6-BA)培养的蛹虫草设置为实验组,分析2种激素在蛹虫草发育过程中对子实体中核苷类物质合成的影响。实验结果表明,10种核苷类物质的检测精密度RSD为0.03%~0.51%,稳定性RSD为0.05%~0.53%,保留时间RSD为0.04%~0.92%,重复性RSD为0.15%~1.53%,说明该检测方法是准确可用的;蛹虫草中核苷类物质含量测定结果显示,供试的2种激素未对胞嘧啶、胞苷、次黄嘌呤的合成产生影响;但均能在特定添加剂量下刺激蛹虫草合成对照组不产生的尿嘧啶和腺嘌呤,仅萘乙酸在添加剂量为6 mg/kg时能使蛹虫草在发育中期产生对照组不产生的胸苷;培养基中按照12 mg/kg的比例添加6-BA可以使成熟蛹虫草子实体中的尿苷含量高达215.28 μg/g,按照24 mg/kg的比例添加6-苄氨基嘌呤可使子实体中鸟苷的含量达到158.64 μg/g,显著高于其他实验组;中高剂量6-BA和萘乙酸在培养基中的添加可以有效促进蛹虫草在发育中期大量累积腺苷,使其含量分别达到198.70、179.32 μg/g和129.62、257.22 μg/g;低剂量6-BA和萘乙酸的添加能刺激蛹虫草成熟子实体中虫草素含量分别高达1219.6 μg/g和1348.24 μg/g,显著高于其他组别。结果表明植物激素6-BA和萘乙酸能够调控蛹虫草中核苷类物质的合成。A high performance liquid chromatography (HPLC) method for quantitative determination of cytidine,guanosine,cytosine,uracil,hypoxanthine,adenine,uridine,adenosine,thymidine and cordycepin was established.In the process of artificial cultivation of Cordyceps militaris,Cordyceps militaris cultured without plant hormones was taken as the control group,and Cordyceps militaris with 6 mg/kg,12 mg/kg and 24 mg/kg of NAA and 6-BA were added into the culture medium to set up experimental groups.The effects of the two hormones on the synthesis of nucleosides in the fruiting body during the development were analyzed.The experimental results showed that the precision RSD,stability RSD,retention time RSD and repeatability RSD of 10 nucleoside substances were 0.03%~0.51%,0.05%~0.53%,0.04%~0.92% and 0.15%~1.53% respectively,it indicated that the experimental method was accurate and available.The determination of nucleosides in Cordyceps militaris showed that the two hormones had no effect on the synthesis of cytosine,cytidine and hypoxanthine.However,both hormones can stimulate Cordyceps militaris to synthesize uracil and adenine,which are not produced by the control group.Only 6 mg/kg NAA could make Cordyceps militaris produce thymidine which was not produced in the control group.Adding 6-BA at the ratio of 12 mg/kg in culture medium could make uridine content in mature Cordyceps militaris reached to 215.28 μg/g,and adding 6-BA at the ratio of 24 mg/kg could make guanosine content in fruiting body reached to 158.64 μg/g,which was significantly higher than other experimental groups.Cordyceps militaris in the medium and high doses group of 6-BA and NAA could effectively accumulate adenosine in the middle development stage,and the content reached to 198.70 μg/g,179.32 μg/g and 129.62 μg/g,257.22 μg/g respectively.The addition of low-dose 6-BA and NAA made the cordycepin content in mature fruiting bodies to be up to 1219.6 μg/g and 1348.24 μg/g respectively,which was significantly higher than other groups.The experiment re
