摘 要:以巨大口蘑为研究对象,采用水提醇沉经典方法及单因素试验,以液料比、超声时间、提取次数3个影响因素作为自变量,以巨大口蘑多糖得率为响应值,设计三因素三水平的响应面试验优化多糖的提取工艺并评价其体外抗氧化活性。结果表明,巨大口蘑多糖最佳提取条件为:液料比27∶1(mL/g),超声时间40 min,超声提取次数2次。抗氧化活性试验表明巨大口蘑多糖对DPPH和ABTS+自由基的最高清除率可达80%。优化的巨大口蘑多糖提取工艺合理可行,巨大口蘑多糖具有较强的抗氧化活性。With Tricholoma giganteum as the research object,the typical method of water extraction-alcohol precipitation and single factor test are adopted,with liquid-solid ratio,ultrasonic time and extraction times as the independent variables,and the yield of polysaccharides from Tricholoma giganteum as the response value,a response surface test with three factors and three levels is designed to optimize the extraction process of polysaccharides and evaluate the antioxidant activity in vitro.The results show that the optimal extraction conditions for polysaccharides from Tricholoma giganteum are as follows:the ratio of liquid to solid is 27∶1(mL/g),the ultrasonic extraction time is 40 min,the ultrasonic extraction times is 2 times.The antioxidant activity test shows that the highest scavenging rate of polysaccharides from Tricholoma giganteum on DPPH and ABTS+free radicals could reach 80%.The optimized extraction technology of polysaccharides from Tricholoma giganteum is reasonable and feasible,and the polysaccharides from Tricholoma giganteum have strong antioxidant activity.
