摘  要:以玉米须为原料,采用碱提酸沉法提取玉米须芦丁,在单因素实验的基础上,分别通过正交实验和响应面法考察了料液比、水浴温度、水浴时间等因素对玉米须芦丁提取率的影响。结果表明:两种方法分析各因素对玉米须芦丁提取率的影响结果基本一致,即料液比>水浴温度>水浴时间。正交实验确定的最优工艺为:料液比1∶15(g∶mL)、水浴温度80℃、水浴时间40 min,玉米须芦丁提取率为5.8945%;响应面法确定的最优工艺为:料液比1∶17(g∶mL)、水浴温度85℃、水浴时间40 min,玉米须芦丁提取率为6.5328%。在响应面法确定的最优工艺条件下的玉米须芦丁提取率优于正交实验确定的最优工艺条件,因此,选择响应面法确定的最优工艺提取玉米须芦丁。Using corn silk as a raw material,we extracted rutin from corn silk by an alkali extraction and acid precipitation method.On the basis of single-factor experiments,we investigated the effects of solid-liquid ratio,water bath temperature,and water bath time on the extraction rate of rutin from corn silk by orthogonal experiments and response surface methodologies,respectively.The results show that the effects of the factors obtained by the two methods on the extraction rate of rutin from corn silk are basically the same,that is,solid-liquid ratio>water bath temperature>water bath time.The optimal extraction conditions by orthogonal experiments are as follows:the solid-liquid ratio of 1∶15(g∶mL),the water bath temperature of 80℃,and the water bath time of 40 min,and the extraction rate of rutin from corn silk is 5.8945%;while the optimal extraction conditions by response surface methodologies are as follows:the solid-liquid ratio of 1∶17(g∶mL),the water bath temperature of 85℃,and the water bath time of 40 min,and the extraction rate of rutin from corn silk is 6.5328%.The extraction rate of rutin from corn silk under the optimized conditions by response surface methodologies is higher than that by orthogonal experiments,therefore the optimal extraction process of rutin from corn silk is determined according to response surface methodologies.
