摘　　要：通过诱变育种获得生物发酵法生产7-ADCA的菌株。将CPC(头孢菌素)产生菌Cephalosporium acremonium B-1822-S-04菌株,通过紫外线-盐酸羟胺复合诱变、菌株筛选,获得一株DAOC(脱乙酰氧头孢菌素)产量突变株D-G3-B-2001,摇瓶发酵DAOC产量为2894μg/mL,50 L发酵罐发酵DAOC产量高达6289μg/mL。将发酵液酸化、过滤处理后,加入酯酶水解,再经DM700树脂吸附、解析处理,得到最终DAOC解析液纯度94%,向DAOC溶液加入CPC酰化酶,反应生成7-ADCA(7-氨基去乙酰氧基头孢烷酸),其纯度达到99.3%以上。该实验筛选出的菌株可为生物发酵法生产7-ADCA提供优质菌株资源和发酵工艺指导。A strain producing 7-ADCA by biological fermentation was obtained by mutation breeding.A mutant strain D-G3-B-2001 with DAOC yield was obtained from CPC producing strain cephalosporium acremonium b-1822-s-04 by UV-hydroxylamine hydrochloride compound mutagenesis and strain screening.The yield of DAOC fermented by mutant strain in shake flask was 2894μg/m L,and the yield of DAOC fermented by 50 L fermentor was as high as 6289μg/m L.The fermentation broth was acidified,filtered,and hydrolyzed by esterase,and then adsorbed and resolved by DM700 resin;the purity of the final DAOC solution was 94%.The CPC acylase was added to the DAOC solution to produce 7-ADCA,and its purity was over 99.3%.The strains screened in this experiment can provides high-quality strain resources and fermentation process guidance for the production of 7-ADCA by biological fermentation.
