摘 要:采用经典乳酸菌筛选法从黑山羊小肠和直肠中分离、纯化乳酸菌,采用形态观察、生理生化试验和16S rDNA序列分析对筛选乳酸菌进行鉴定,并对其生长性能、产酸性能、耐受性及抑菌性能进行测定。结果表明,共分离到32株乳酸菌,可归为5类,从中选取5株代表菌株进行鉴定,其中2株为植物乳杆菌(Lactobacillus plantarum),2株为发酵乳杆菌(Lactobacillus fermentum),1株为融合魏斯氏菌(Weissella confusa)。其中,菌株HCA5-3长势较好;菌株HCS4-1产酸速率较快,菌株HCA6-2产酸能力较强;菌株HCS4-3耐酸性(pH 3.5)较好;菌株HCA6-2、HCA5-3和HCS4-3耐高温(45℃)能力较强,菌株HCA6-2、HCS4-3耐低温(10℃)能力较强;5株代表菌株耐盐性(NaCl 6.5%)能较好,对致病菌均具有较好的抑制作用。Lactic acid bacteria were isolated and purified from small intestine and rectum of Hainan black goats by classical lactic acid bacteria screening method,and identified by morphologic observation,physiological and biochemical tests and 16S rDNA sequence analysis.At the same time,the performance of growth,acid production,tolerance and inhibition were determined.The results showed that a total of 32 strains of lactic acid bacteria were isolated,which could be classified into 5 groups.Among them,5 representative strains were selected for identification.After identification,2 strains were Lactobacillus plantarum,2 strains were Lactobacillus fermentum,and 1 strain was Weissella confusa.Among them,the strain HCA5-3 had stronger growth ability.The strain HCS4-1 had higher acid production rate,and the strain HCA6-2 had stronger acid production ability.The strain HCS4-3 had better acid resistance(pH 3.5),the strains HCA6-2,HCA5-3 and HCS4-3 had stronger high temperature tolerance(45℃),and the strains HCA6-2 and HCS4-3 had stronger low temperature tolerance(10℃).All 5 representative strains had better salt tolerance(NaCl 6.5%)and antibacterial activity on pathogens.
