摘  要:从海泥中筛选到了一株高产低温葡萄糖氧化酶(Glucose oxidase,GOD)的担子菌(Basidioascus sp.)酵母菌株(31号),在前期单因素优化实验(单因素优化结果为:蔗糖4%、酵母粉0.3%、NaNO_(3)0.4%、无机盐添加量(KH_(2)PO_(4)0.02%、KCl 0.10%、MgSO_(4)·7H_(2)O 0.10%)、初始pH7.0、装液量125mL/250mL、接种量2%、发酵转速200r/min、发酵温度20℃,酶活力达到435.37U/mL)的基础上,对其进行PlackettBurman、Box-Behnken等相关实验,以进一步提高31号菌株产GOD酶活性,结果表明,GOD酶活性显著提高。Plackett-Burman实验确定了发酵温度、蔗糖添加量和接种量是影响31号菌株产酶的重要因素。最陡爬坡实验及响应面实验联合优化,确定了三个最显著影响因素的最佳培养条件为发酵温度26℃、蔗糖3.92%和接种量3.58%。根据实际情况,进行适当调整,确定31号菌株产GOD酶的最佳条件,在最优发酵条件下,菌株产GOD酶活力达503.22U/mL,与优化前酶活435.37U/mL相比,优化后GOD酶活提高了15.58%。Strain 31 originated from sea mud of the Bohai Sea in Dalian.In order to improve GOD enzyme activity of strain 31,based on single factor optimization in the early stage,Plackett-Burman,steepest climbing and Box-Behnken experiments were performed to select the best conditions on enzyme activity.The results showed GOD enzyme activity is significantly improved.The Plackett-Burman experiment determined that the fermentation temperature,the amount of sucrose and inoculum are important factors that affect the enzyme production of strain 31.The steepest climbing experiment and response surface experiment were combined in selecting the optimal culture conditions.The best conditions for the three most significant influencing factors were:the fermentation temperature 26℃,sucrose amount 3.92%,and the inoculum amount 3.58%.According to the actual situation,appropriate adjustments were made to select the optimal conditions for low-temperature GOD enzyme production of strain 31.Under the optimal fermentation conditions,the low-temperature GOD enzyme activity reached to 503.22U/mL.Compared with 435.37 U/mL before the optimization,the GOD enzyme activity increased by 15.58%.
