摘  要:研究采用透明圈法筛选1株耐高温木聚糖酶产生菌TP-1。通过菌落特征观察、显微镜观察和ITS序列分析,鉴定为杜邦嗜热菌。结果显示,经酶学性质初步研究杜邦嗜热菌TP-1所产的木聚糖酶的最佳作用温度为60℃,最佳反应pH值为5,最适条件下测得酶活为75.96 U/g。pH值4、60℃条件下保温2 h,该酶仍可保持较高活性。研究表明,Fe^(2+)、Ba^(2+)、Mn^(2+)对木聚糖酶酶活具有明显的提升作用,Co^(2+)能够降低酶活。该酶在饲料加工行业具有较好应用前景。The study used the transparent circle method to screen a high-temperature-tolerant xylanase-producing strain TP-1. It was identified as Thermomyces dupontii through the observation of colony characteristics, microscope observation and ITS sequence analysis. The result showed that the optimal temperature of the xylanase produced by Thermomyces dupontii TP-1 was 60 ℃, the optimal reaction pH value was 5, and the enzyme activity was 75.96 U/g under the optimal conditions. Incubate at p H value 4 and 60 ℃ for 2 h, the enzyme can still maintain high activity. The experiment indicates that Fe^(2+), Ba^(2+), and Mn^(2+)can significantly improve the enzyme activity of xylanase, and Co^(2+)can reduce the enzyme activity. The enzyme has good application prospects in feed processingindustry.
