摘 要:为筛选出产脂肪酶的菌株,从油茶枯饼发酵液中取样,用中性红油脂培养基进行初筛,改进铜皂—分光光度法测定酶活力进行复筛,获得1株产脂肪酶活性较强的菌株ck-01。结合形态学观察、生理生化试验、16S rDNA序列鉴定和系统发育分析等方法将其鉴定为蜡状芽孢杆菌。对菌株ck-01的酶学性质的研究结果表明,该菌株所产脂肪酶在35℃和pH值7.0的条件下,酶活力为3.35U/mL,Ca^(2+)、Mg^(2+)对酶活有促进作用,Fe^(2+)、Zn^(2+)对酶活有抑制作用,EDTA和SDS可使酶失活。In order to screen the lipase producing strain,the high lipase activity producing strains were isolated from fermented liquid of oil camellia cake,through conducting primary screen using neutral red oil medium and re-screening by determining the enzyme activity using the improved copper soap-spectropho-tometer method.The highest lipase activity producing strain,named ck-01,was further analyzed.Strain ck-01 was identified as bacillus cereus by morphological observation,physiological and biochemical experiment,16S rDNA sequence analysis and phylogenetic analysis.In addition,the enzymatic properties analysis of Strain ck-01 showed that the lipase activity was 3.35 U/mL under the condition of 35℃ and pH 7.0.The lipase activity of Strain ck-01 was promoted by Ca^(2+)and Mg^(2+),but it was inhibited by Fe^(2+)and Zn^(2+).It appeared inactivation after being treated by EDTA and SDS.
