摘  要:采用传统乙醇回流法提取油莎草块茎中总生物碱,研究乙醇体积分数、料液比、提取时间、提取温度对油莎草块茎总生物碱提取得率的影响。在单因素试验基础上,通过L9(34)正交试验优化油莎草块茎总生物碱提取工艺,采用pH示差法测定莎草块茎中总生物碱含量。结果表明,油莎草块茎总生物碱最优提取工艺为乙醇体积分数60%、料液比1︰13 g/mL、提取时间3 h、提取温度70℃。在此条件下,油莎草总生物碱提取得率为1.2250 mg/g。确定采用p H示差法测定莎草块茎中总生物碱含量。The optimum extraction process of total alkaloids from Cyperus esculentus L.tubers(CELt)was gained by using traditional ethanol reflux method while the effects of ethanol concentration,material-liquid ratio,extraction time and extraction temperature on extraction rate of total alkaloids from CELt were observed.On the basis of single factor experiment,the total alkaloids extraction process was optimized by orthogonal experiment.The content of total alkaloids in CELt was determined by pH differential method.The results showed that the optimal extraction process of total alkaloids from CELt were as follows:ethanol concentration 60%,ratio of material to liquid was of 1︰13 g/mL,extraction time 3 h and extraction temperature 70℃.Under the optimal condition,the yield of total alkaloids was 1.2250 mg/g.The content of total alkaloids in CELt was determined by pH differential method.
