摘  要:利用大孔树脂分离纯化桤叶唐棣花色苷,通过吸附—解吸试验确定纯化的最佳工艺条件。结果显示:将pH=2.0、花色苷质量浓度为0.100 g/L的粗提液按2柱体积/h通入XDA-8大孔树脂柱,再用体积分数为30%、50%、70%、90%乙醇以3柱体积/h各冲洗4柱体积,花色苷纯度从2.13%增加到39.55%,提高了18.57倍。结合UPLC-MS对纯化物进行鉴定分析,确定主要含矢车菊素-3-O-葡萄糖苷、飞燕草素-3-葡萄糖苷和锦葵素-3-葡萄糖苷3种成分。经抗氧化和抑菌试验,发现纯化物具有还原性,能有效清除1,1-二苯基-2-三硝基苯肼(DPPH)自由基和羟自由基(·OH),清除率与质量浓度呈正相关;纯化物对3种供试菌具有不同的抑制效果,其由大到小为金黄色葡萄球菌、大肠杆菌、枯草芽孢杆菌,且质量浓度越大,抑菌性越强。因此,XDA-8大孔树脂可用于纯化桤叶唐棣花色苷,纯化物具有抗氧化性,且对细菌可以起到抑制作用。The anthocyanins of Amelanchier alnifolia were isolated and purified using macroporous resin.Basing on the study of the loading and the eluting conditions,the optimum purification process parameters were determined.The optimum conditions for separation and purification were as follows:anthocyanins concentration was 0.100 g/L,pH was 2.0,loading flow rate was 2 BV/h,elution flow rate was 3 BV/h,and the elution volume was 4 BV with 30%,50%,70%,90%ethanol respectively.Under the above conditions,the purity of anthocyanins from A.alnifolia increased by 18.57 folds from 2.13%to 39.55%.The structure of the purified anthocyanins was identified by using UPLC-MS,and three anthocyanin components were identified as cyanidin-3-o-glucoside,delphinidin-3-glucoside,malvidin-3-glucoside.Through the antioxidant and antibacterial experiments,the purified anthocyanins from A.alnifolia had reducing power and scavenging activity against DPPH·and·OH in a concentration-dependent fashion.The antibacterial activity of purified anthocyanins from A.alnifolia against different strains followed the decreasing order of Staphylococcus aureus,Escherichia coli and Bacillus subtilis,and was positively correlated with concentration.It was concluded that XAD-8 macroporous resin could be used to purify the anthocyanins from A.alnifolia,and the purified anthocyanins had good antioxidant activity and inhibitory effect on bacteria.
