Optimization of Extraction Process of Chlorogenic Acid from Lonicera japonica with Cellulase and Its Antioxidative Activity

摘  要:利用纤维素酶法提取金银花中绿原酸并优化提取工艺;以维生素C为阳性对照,利用铁离子还原法和羟自由基清除试验确定所得金银花绿原酸提取液的体外抗氧化活性。结果表明,各因素对金银花绿原酸提取率的影响顺序为纤维素酶添加量>提取温度>提取时间>pH值,在纤维素酶添加量0.4%、提取温度50℃、提取时间50 min、pH4.5的条件下,金银花绿原酸提取率最高,达到40.10 mg/g。所得绿原酸提取液有较好的体外抗氧化活性,铁离子还原能力高于维生素C,0.5 mg/mL的绿原酸提取液对羟自由基清除率达44.51%,高于相同浓度维生素C的羟自由基清除率(28.48%)。This study optimized the extraction procedure of chlorogenic acid from Lonicerajaponica with cellulase,and measured its antioxidant activity in vitro with V_(c) as positive control via iron reduction method and hydroxyl radical scavenging experiment.The weight of the factors examined was as follows;cellulase dosage>extraction temperature>extraction time>pH value,and the optimal parameters were cellulase dosage of 0.4%,extraction temperature of 50℃,extraction time of 50 min,and pH4.5.The chlorogenic acid yield from Lonicera japonica was 40.10 mg/g.The results showed that the chlorogenic acid extract had antioxidant activity in vitro,and its ferric ion reducing capacity was higher than that of V_(c) The hydroxyl radical scavenging rate of 0.5 mg/mL extract was 44.51%,which was higher than that of V_(c)(28.48%)at the same concentration.
