摘 要:鱼皮含有丰富的胶原蛋白,为优化胶原蛋白提取方法,本文以鲫鱼鱼皮作为提取材料,使用双酶法和超声-双酶法提取鲫鱼皮胶原蛋白,以料液比、风味蛋白酶加入量、碱性蛋白酶加入量和酶解温度为实验因素,探究最佳提取条件。结果表明,使用超声-双酶法在超声60 min,料液比为1∶10(g∶mL),2种蛋白酶加入量为4 000 U/g,酶解温度45 ℃的反应条件下,胶原蛋白提取效果最好,得到的提取率为14.70%。使用双酶法在料液比例为1∶15(g∶mL),2种蛋白酶加入量为4 000 U/g,酶解温度50 ℃的反应条件下,胶原蛋白提取效果最好,得到的提取率为10.53%。相较于双酶法,超声-双酶法提取鲫鱼鱼皮胶原蛋白效果更佳,在更低的料液比和酶解温度条件下提取率更高。Fish skin is rich in collagen,in order to optimize the collagen extraction method,the double enzyme method and ultrasonic-double enzymatic method were used to extract collagen from crucian carp (Carassius auratus) skin,which was used as raw material.Solid-liquid ratio,flavor protease content,alkaline protease content and enzymatic hydrolysis temperature were taken as experimental factors,explore the best extraction conditions.The results showed that under the conditions of ultrasonic time 60 min,solid-liquid ratio 1∶10(g∶mL),two kinds of proteases addition 4 000 U/g and enzymolysis temperature 45 ℃,collagen extract by ultrasonic-dual enzymatic method is the best,the extraction rate of collagen was 14.70%.Solid-liquid ratio 1∶15(g∶mL),two kinds of proteases addition 4 000 U/g,and enzymolysis temperature 50 ℃,the extraction rate of collagen was 10.53%.Compared to the double enzymatic method,the ultrasonic-double enzyme method was more effective in extracting collagen from crucian carp skin,and the extraction rate was higher at lower solid-liquid ratio and enzymolysis temperature.
