摘  要:试验旨在探究铁皮石斛中SOD的提取纯化方法和在酸奶中的运用。用热变性的方式提取1年生铁皮石斛鲜条中的SOD,纯化的方式是硫酸铵分级沉淀法;以邻苯三酚为质料,测定SOD对邻苯三酚自动氧化速度的影响。将SOD应用于发酵酸奶中,研究其在食品中的应用。SOD酶活性为8.72 U/mL,用硫酸铵对其进行分级沉淀后的SOD酶活性为10.22 U/mL,酶活性比提纯前有显著上升;提纯后SOD酶活性极高温度为70℃;在酸奶中,发酵之前加SOD之后的24 h酶活性较高。试验对铁皮石斛中SOD的初步提取、纯化及应用进行研究,为食品中SOD的开发提供参考。To explore the extraction and purification method of SOD from Dendrobium officinale and its application in yoghurt,SOD was extracted from fresh strips of annual Dendrobium officinale by thermal denaturation method and the purification method was ammonium sulfate fractionated precipitation method.The effect of superoxide dismutase(SOD)on the autooxidation rate of pyrogallol was studied.SOD was used in fermented yogurt to study its adhibition in food.SOD enzyme liveness was 8.72 U/mL,and SOD enzyme liveness was 10.22 U/mL after fractional precipitation with ammonium sulfate.The activity of SOD enzyme compared with before purification was significantly increased.The highest temperature for SOD enzyme activity after purification was 70 ℃.SOD activity increased after 24 h in yogurt with SOD enzyme added before fermentation.The preliminary extraction,purification and application of SOD in Dendrobium officinale were studied,which provided reference for the development of SOD enzyme in food.
