摘 要:灵芝多糖(Ganoderma lucidum polysaccharide,GLP)具有良好的生物学活性,在医药、食品、化妆品等领域有广泛应用.本文以碱溶性灵芝多糖为原料制备羧甲基灵芝多糖(Carboxylmethylated GLP,CM-GLP),并考察其溶解性、红外光谱和体外抗氧化能力.结果显示,当氯乙酸用量3.8 g,NaOH浓度4.5 mol/L,醚化温度65 oC时,CM-GLP羧甲基取代度较高;经羧甲基化后,GLP水溶性有很大改善;红外光谱显示,样品中含有羧基的伸缩振动峰,说明GLP羧甲基化成功;在0~10 mg/mL范围内,CM-GLP的清除羟基自由基能力和还原力与CM-GLP浓度和取代度之间均呈正相关,说明通过羧甲基化可提高碱溶性GLP的溶解性和抗氧化性.Ganoderma lucidum polysaccharide(GLP)has good biological activity and is widely used in medicine,food,cosmetics and other fields.In this work,carboxymethylated GLP(CM-GLP)was prepared from alkali soluble GLP through carboxymethylation.In addition,the water solubility,infrared spectrum(IR)and in vitro antioxidant capacity of CM-GLP were also investigated.The results showed that when the dosage of chloroacetic acid was 3.8 g,the concentration of NaOH was 4.5 mol/L,the etherification temperature was 65 oC,a high degree of carboxymethyl substitution of CM-GLPwas obtained.After carboxymethylation,the water solubility of CM-GLPwas greatly improved.IR result showed that the sample contained the stretching vibration peak of carboxyl group,indicating that the carboxymethylation of GLP was successful.In the range of 0~10 mg/mL,the hydroxyl radical scavenging ability and reducing power of CM-GLP were positively correlated with the concentration and degree of substitution of CM-GLP,indicating that carboxymethylation can improve the solubility and antioxidant capacity of alkali soluble GLP.
