摘 要:以青钱柳叶片提取多糖后的残渣为原料,对酸碱法、超声辅助碱法、微波辅助法、双酶法和超声辅助酶法提取的不溶性膳食纤维(insoluble dietary fiber,IDF)的理化性质和抗氧化活性进行比较研究。红外光谱显示,5种方法提取的IDF均具有纤维素类多糖的特征吸收峰,且峰形相似。酸碱法和超声辅助碱法提取的IDF具有较高的膨胀率;超声辅助碱法提取的IDF具有良好的持水力和持油力。不同方法提取的IDF抗氧化活性之间存在差异,综合DPPH自由基清除率、羟基自由基清除率和铁离子还原/抗氧化能力,微波辅助法提取的IDF具有较高的抗氧化活性。Five methods,includingacid-alkali,ultrasound-assisted alkali,microwave-assisted,double-enzyme,and ultrasound-assisted enzyme extraction,were compared to prepare the insoluble dietary fiber(IDF)from the residue of Cyclocarya paliurus leaves after polysaccharide extraction and the physicochemical properties and antioxidant activity of IDF were measured.The infrared spectrum showed that the IDF extracted by the five methods all had the characteristic absorption peak of cellulose polysaccharide,and the peak shape was similar.The IDF extracted by the acid-alkali method and the ultrasound-assisted alkali method had higher expansion rates,and that extracted by the ultrasound-assisted alkali method had a better water holding capacity and oil holding capacity.The IDF extracted by different methods had variedantioxidant activities.Based on the DPPH free radical scavenging rate,hydroxyl free radical scavenging rate,and ferric ion reducing antioxidant power,the IDF extracted by microwave-assisted method had a higher antioxidant activity.
