摘 要:芽孢杆菌在农作物生物防治方面具有重大潜力。本研究以实验室前期分离的暹罗芽孢杆菌X7为材料,采用DEAE-Sephadex A-25层析、Sephadex G-75凝胶层析以及变性聚丙烯酰胺凝胶电泳(SDS-PAGE)相结合的方式分离纯化具有拮抗作用的抗菌肽,并对抗菌肽的抑菌特性和稳定性进行分析。结果表明,暹罗芽孢杆菌X7能产生小分子抑菌物质,经纯化后可获得单一条带,相对分子质量约为20.1 kDa。抑菌物质具有一定的温度稳定性、酸碱稳定性、紫外线稳定性、光照稳定性,对蛋白酶、有机试剂不敏感。研究结果可为暹罗芽孢杆菌X7抗菌肽在农作物生物防治中的应用提供理论依据。Bacillus has great potential in crop biological control.In this study,the Bacillus siamese X7 isolated previously in our laboratory was used as material to separate and purify antibacterial peptides with antagonistic effects using DEAE-Sephadex A-25 chromatography,Sephadex G-75 gel chromatography and denaturing polyacrylamide gel electrophoresis(SDS-PAGE).The antibacterial properties and stability of the antibacterial peptides were also analyzed.The results showed that B.siamese X7 could produce small molecule antibacterial substances,and a single band could be obtained after purification with relative molecular mass as about 20.1 kDa.The antibacterial substances had certain light stability,acid-base stability,ultraviolet stability and light stability,and were not sensitive to proteases and organic reagents.These results could provide theoretical bases for application of B.siamensis X7 antimicrobial peptides in crop biological control.
