摘  要:以鱿鱼内脏为原料,以提取时间、提取温度、料液比为3个考察因素,在单因素试验基础上,通过响应面法优化鱿鱼内脏中章鱼胺的提取工艺,并探究章鱼胺体外抗氧化活性。通过高效液相色谱对鱿鱼内脏中章鱼胺进行定性定量分析。结果显示,章鱼胺最佳提取工艺参数:以5%三氯乙酸作为提取剂,提取时间2 h,提取温度34℃,料液比1∶5.2(g/mL),此条件下鱿鱼内脏中章鱼胺得率为6.51 mg/g,与预测值6.25 mg/g较为接近。抗氧化试验研究表明,提取的章鱼胺具有明显的抗氧化活性。Using squid viscera as raw material,the influence of extraction time,extraction temperature,and material-to-liquid ratio on the extraction of octopamine from squid viscera was first investigated by single factor experiment,and then the extraction process was optimized by response surface methodology.Moreover,the in vitro antioxidant activity of octopamine was tested.High performance liquid chromatography was performed for qualitative and quantitative analysis of the octopamine in squid viscera.The results showed that the optimal extraction parameters were 5%trichloroacetic acid as extraction solvent,material-to-liquid ratio of 1∶5.2(g/mL),and extraction for 2 h at 34℃.Under these conditions,octopamine yield reached 6.51 mg/g,which was close to the predicted value of 6.25 mg/g.In addition,the extracted octopamine had obvious antioxidant activity.
