摘  要:目的比较干姜、桂枝及其药对挥发性成分的相对百分含量,探讨干姜、桂枝药对配伍的意义。方法采用顶空固相微萃取法(HS-SPME)与气相色谱-质谱(GC-MS)联用法,分析干姜、桂枝及其药对的挥发性成分。通过单因素考察法确定最佳萃取条件,采用面积归一化法计算各挥发性成分的相对百分含量,并进行主成分分析(PCA)。结果通过单因素考察法确定样品的最佳萃取条件为:进样量0.8 g,萃取温度60℃,萃取时间20 min,解析时间3 min。从干姜、桂枝及其药对中分别鉴定出43、35、38种挥发性成分,各占总挥发性成分含量的89.84%、80.69%和98.36%。干姜、桂枝及其药对共有的挥发性成分有9种,但共有成分的含量有所不同。三者的PCA综合得分具有明显的差异,药对的挥发性成分的PCA综合得分最高。结论干姜和桂枝配伍前后,单味药与药对的挥发性成分存在差异,且产生了新的挥发性成分。干姜、桂枝药对PCA综合得分最高,从挥发性成分变化推测,干姜和桂枝配伍后具有一定的协同增效的作用。Objective To compare the relative percentage of volatile components in Zingiberis rhizome,Cinnamomi ramulus and the combination of these two(the drug pair),and to explore the compatibility of Zingiberis rhizome and Cinnamomi ramulus.Methods Headspace solid phase microextraction(HS-SPME)and gas chromatrometry(GC)mass chromatrometry(MS)were used to analyse the volatile components of Zingiberis rhizome,Cinnamomi ramulus and the drug pair.The optimal extraction conditions were determined by single factor method,and the relative percentage of volatile components was calculated by area normalization method,and principal component analysis(PCA)was performed.Results The optimal extraction conditions determined by single factor method were as follows:sample weight to be tested was 0.8 g,the extraction temperature was 60℃,with an extraction duration of 20 min,and an analytical time of 3 min.Forty-three volatile components were identified from Zingiberis rhizome,while 35 from Cinnamomi ramulus,and 38 from the drug pair,which accounted for 89.84%,80.64%,and 98.36%of the total volatile components respectively.There were 9 common volatile components in Zingiberis rhizome,Cinnamomi ramulus and the drug pair,but the contents of common volatile components were different.In addition,the PCA scores of the three groups were significantly different,and the comprehensive scores of drug pair for volatile components were the highest.Conclusion Before and after the combination of Zingiberis rhizome and Cinnamomi ramulus,the volatile components of single herb and drug pair were different,and new volatile components were produced.The results of PCA showed that the comprehensive score of drug pair was the highest,and it could be inferred from the changes of volatile components that,the combination of Zingiberis rhizome and Cinnamomi ramulus had synergistic effect.
