摘  要:本文开展了产脂肪酶菌株的筛选鉴定及响应面法优化发酵工艺试验,确定得到的高产脂肪酶菌株为Youzh-1,对其进行16S rRNA测序分析,确定其为荧光假单胞菌,在单因素试验的基础上采用响应面法对Youzh-1的发酵工艺进行进一步优化设计,得到最佳发酵工艺为葡萄糖添加量18.825 g/L、牛肉膏添加量25.98 g/L、温度41℃、pH值6。在此条件下测得脂肪酶活力为79.87 U/mL,较理论值87.19 U/mL减少了8.39%,证明模型可靠。In this paper,the screening and identification of lipase-producing strain and the optimization of fermentation process by response surface methodology were researched.The obtained high lipase-producing strain was Youzh-1,which was determined to be Pseudomonas fluorescens by 16S rRNA sequencing analysis.On the basis of the single factor experiment,the response surface methodology was used to further optimize the fermentation process of Youzh-1.The optimum fermentation process was obtained as follows:the addition of glucose was 18.825 g/L,the addition of beef extract was 25.98 g/L,the temperature was 41℃,and the pH value was 6.Under these conditions,the lipase activity was measured to be 79.87 U/mL,which was 8.39%lower than the theoretical value(87.19 U/mL),which proved that the model was reliable.
