摘  要:本文主要通过建立荧光PCR法实现高效鉴别大西洋鲑鱼成分的目的。以鲑形目8种鱼类的COⅠ基因序列作为目标基因来与大西洋鲑鱼基因作比对,以差异序列作为其特异性引物,对鉴定大西洋鲑鱼源性成分的方法——荧光定量PCR法进行建立与完善。实验选用同目的 6种近源物种和45种非近源物种进行特异性检验,能够对大西洋鲑鱼源性成分的特异性进行有效识别,且具有较高灵敏度,仅0.1 ng/μL大西洋鲑鱼DNA即可被检出。实验对市售的5种鱼松,共计32个受测样本进行有效检测,证实此方法对鱼松中的大西洋鲑鱼成分鉴别具有较高的灵敏度和特异性,可广泛应用于鱼松中大西洋鲑鱼源性成分的识别与鉴定。In this study, a fluorescence quantitative PCR method was established to identify the components of Atlantic salmon floss. Comparison of CO Ⅰ gene sequences of 8 salmoniforms were target genes, and the differential sequences were used as the specific primers to establish and improve the real-time PCR method for identifying the source components of Atlantic salmon. The method was specific for 6 near-source species and 45 non-near-source species of the same order, which could effectively identify the specificity of components derived from Atlantic salmon and had a high sensitivity. It could be detected with 0.1 ng/μL Atlantic salmon DNA. The experiment carried out the effective detection on 5 kinds of fish floss in the market, a total of 32 tested samples, which confirmed its high sensitivity and specificity for identifying the Atlantic salmon components in fish floss and could be widely applied.
