摘 要:目的:探究茶多糖-茶多酚混合功能成分(tea extract rich in polysaccharides and polyphenols,TEPP)是否能够改善D-半乳糖诱导的小鼠肠道氧化应激反应,同时探究其抗氧化机制。方法:小鼠腹腔注射质量分数10%D-半乳糖8周建立氧化应激模型,造模成功后,正常组与模型组灌胃蒸馏水,阳性组灌胃200 mg/kg m_(b)还原型谷胱甘肽,TEPP低、中、高剂量组分别灌胃40、100、250 mg/kg m_(b)的TEPP,茶多酚组灌胃50 mg/kg m_(b)的茶多酚。用酶联免疫吸附试验试剂盒检测各组小鼠肠组织的总抗氧化能力、活性氧质量浓度、谷胱甘肽还原酶活力、血红素加氧酶活力,通过实时荧光定量聚合酶链式反应法(quantitative real-time polymerase chain reaction,qPCR)检测各组肠组织Toll样受体4(toll-like receptors,TLR4)、p38、核因子NF-E2相关因子(nuclear factor-erythroid 2-related factor 2,Nrf2)、谷胱甘肽还原酶、血红素加氧酶的mRNA表达量。结果:与模型组相比,TEPP高剂量组的总抗氧化能力上升了约2.2倍,谷胱甘肽还原酶、血红素加氧酶活力分别上升了2.8、1.7倍,活性氧浓度降低了72.8%,TLR4、p38的mRNA表达量分别降低了39.1%、82.4%,Nrf2、谷胱甘肽还原酶、血红素加氧酶的mRNA表达量分别升高了83.6%、8.3倍、96.3%,这些指标变化都与模型组存在显著差异(P<0.05)。结论:TEPP能够通过TLR4/p38丝裂原活化蛋白激酶(p38 mitogen-activated protein kinase,p38 MAPK)/Nrf2通路改善D-半乳糖诱导的小鼠肠道的氧化应激反应。Objective:To evaluate whether a tea extract rich in polysaccharides and polyphenols(TEPP)can improve the intestinal response to oxidative stress induced by D-galactose in mice,and to explore its antioxidant mechanism.Methods:Mice were injected intraperitoneally with 10%D-galactose for eight weeks to establish an oxidative stress model.After the model was successfully established,the normal and model groups were gavaged with distilled water,the positive group was gavaged with 200 mg/kg m_(b) of reduced glutathione,the low-,medium-and high-dose TEPP groups were respectively administered with 40,100 and 250 mg/kg m_(b) of TEPP,and the tea polyphenol group was administered with 50 mg/kg m_(b) of tea polyphenols.Enzyme-linked immunosorbent assay kits were used to detect the total antioxidant capacity(T-AOC),reactive oxygen species(ROS)content,glutathione reductase(GR)activity,heme oxygenase(HO)activity in the intestinal tissues of the mice in each group,and the mRNA expression levels of toll-like receptors(TLR4),p38,nuclear factorerythroid 2-related factor 2(Nrf2),GR and HO were detected by fluorescent quantitative real-time polymerase chain reaction(qPCR).Results:Compared with the model group,the T-AOC of the high-dose TEPP group increased by 2.2 times,and the activities of GR and HO increased by 2.8 times and 1.7 times,respectively.The ROS content decreased by 72.8%,the mRNA expression of TLR4 and p38 declined by 39.1%and 82.4%,and the mRNA expression of Nrf2,GR and HO increased by 83.6%,8.3 times and 96.3%,respectively.All of these changes were statistically significant(P<0.05).Conclusion:TEPP can improve the intestinal response of mice to oxidative stress induced by D-galactose through the TLR4/p38 mitogenactivated protein kinase(MAPK)/Nrf2 pathway.
